This gene is a member of the aggrecan/versican proteoglycan family. The encoded protein is an integral part of the extracellular matrix in cartilagenous tissue and it withstands compression in cartilage. Mutations in this gene may be involved in skeletal dysplasia and spinal degeneration. Multiple alternatively spliced transcript variants that encode different protein isoforms have been observed in this gene. (provided by RefSeq)
Aggrecan (AGC1) is a multimodular molecule, encoded by the gene mapped on human chromosome 15q26.1. It is a large aggregating chondroitin sulfate proteoglycan of the human cartilage. The core protein of this multimodular molecule contains three globular domains (G1, G2 and G3) and a large extended region (CS) between G2 and G3 for glycosaminoglycan chain attachment.

AGC1 (NP_037359, 56 a.a. ~ 155 a.a) partial recombinant protein with GST tag. MW of the GST tag alone is 26 KDa.

Aggrecan (AGC1) is required for proper functioning of articular cartilage. The encoded protein plays an essential role in chondroskeletal morphogenesis during development. AGC1 facilitates chondrocyte-chondrocyte and chondrocyte-matrix interactions through its ability to bind hyaluronan. Mutations in aggrecan results in autosomal dominant short stature with accelerated skeletal maturation. In addition, genetic variation in the gene is also associated with the development of spondyloepiphyseal dysplasia.
