DARPP32 is a phosphoprotein with a molecular mass of 32kDa. It is regulated by dopamine (DA) and cAMP and is associated with dopaminoceptive neurons bearing D-1 receptors in the basal ganglia. DARPP32 is a target for the actions of dopamine.

Dopamine D1 receptor stimulation enhances cAMP formation, resulting in the phosphorylation of DARPP32. When phosphorylated on threonine 34, DARPP32 acts as a potent protein phosphatase- inhibitor. This phosphorylation plays an important role in the regulation of dopaminergic neurotransmission. The activity of this protein has an important role in the actions of alcohol, caffeine and Prozac^®.
