The gene SHISA6 (shisa family member 6) encodes a single transmembrane protein that is mainly expressed at hippocampal synapses. It is a member of the Shisa family of proteins and contains the characteristic cysteine-rich motif, a single-pass transmembrane region and a type-II PDZ (PSD95-Dlg1-zo-1)-ligand motif at the C-terminal tail of the intracellular domain.

The gene SHISA6 (shisa family member 6) encodes a directly interacting AMPAR (α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid receptor) auxiliary subunit. Its PDZ (PSD95-Dlg1-zo-1)-ligand motif is found to associate with PSD-95 ((postsynaptic density protein 95), and constrains the mobility of AMPARs in the plasma membrane. This restrains the AMPARs to postsynaptic densities and inhibits their desensitization during synaptic activity.
