Three prime histone mRNA exonuclease 1/exoribonuclease 1 (THEX1, ERI1,) is a 3′ exonuclease that binds to the 3′ end of mammalian histone mRNAs and degrades them. The rapid degradation of histone mRNA by ERI1 occurs upon completion and/or inhibition of DNA replication. ERI1 may also negatively regulate RNA interference via degradation of siRNA.

Synthetic peptide directed towards the C terminal region of human THEX1

Anti-THEX1 polyclonal antibody is used to tag three prime histone mRNA exonuclease 1 for detection and quantitation by Western blotting and in plasma by immunohistochemical (IHC) techniques. It is used as a probe to determine the roles of three prime histone mRNA exonuclease 1 in histone mRNA degradation during phases of DNA replication.

THEX1 contains 1 SAP domain and 1 exonuclease domain. It is an RNA exonuclease that binds to the 3′ end of histone mRNAs and probably degrades them, suggesting that it plays an essential role in histone mRNA decay after replication. It is also able to degrade the 3′ overhangs of short interfering RNAs (siRNAs) in vitro, suggesting a possible role as regulator of RNA interference (RNAi).
